Background & Theory
Caffeine works by blocking adenosine receptors in the central nervous system. Adenosine builds up across the waking day and promotes drowsiness; caffeine is shaped closely enough to occupy the same receptor sites without activating them, so the sleep pressure is masked rather than erased. That is why fatigue often arrives all at once once the drug clears. Absorption from the gut is nearly complete, blood levels peak roughly 30 to 60 minutes after a drink, and clearance happens almost entirely in the liver through the CYP1A2 enzyme.
At ordinary dietary doses elimination follows first-order kinetics, which is what makes the decay curve on this page a plain exponential: the amount left after t hours equals the starting dose times 0.5 raised to the power of t divided by the half-life. This calculator uses a five hour half-life as a middle-of-the-road adult value, but real half-lives range from roughly 2 to 10 hours depending on CYP1A2 genotype, smoking (which shortens it markedly), and oral contraceptives or pregnancy (which can double or triple it). The 400 mg daily figure is the widely cited ceiling for healthy adults; the per-kilogram number used here comes from spreading that same 400 mg across a 70 kg reference adult, giving about 5.7 mg per kilogram. The 50 mg sleep threshold is a practical rule of thumb rather than a clinical cutoff, since bedtime sensitivity varies enormously from person to person.
History
Caffeine was first isolated in 1819 by the German chemist Friedlieb Ferdinand Runge, working at Jena after Johann Wolfgang von Goethe handed him a box of coffee beans and asked what was in them. French chemists reported independent isolations within a few years, and across the nineteenth century the same compound was rediscovered in tea, kola nuts, and cacao under separate names before chemists established the substances were identical. Hermann Emil Fischer worked out the structure of caffeine and its related purines and achieved a laboratory synthesis in the 1890s, part of the body of work recognized by his 1902 Nobel Prize in Chemistry.
Explaining how caffeine actually acts took much longer. For decades the leading account was inhibition of phosphodiesterase, but the concentrations needed for that were far above anything a cup of coffee produces. Research through the late 1970s and 1980s, including work by John Daly and colleagues at the U.S. National Institutes of Health, established antagonism of adenosine receptors as the mechanism that operates at dietary doses. Pharmacokinetic studies over the same period pinned down the roughly five hour average half-life and the wide person-to-person spread that this calculator's decay curve approximates. Regulatory guidance settled later still, with 400 mg per day for healthy adults and 200 mg during pregnancy becoming the common reference points.
